Specificity

Detects human Neurturin in direct ELISAs and Western blots. In direct ELISAs, approximately 80% cross-reactivity with recombinant mouse Neurturin is observed and no cross-reactivity with recombinant mouse (rm) Artemin, recombinant human (rh) Cripto-1, recombinant Drosophila DPP, rhGDNF, rhLefty A, rmLefty-1, rhMIS, rmNodal, rmPersephin, rhTGF-beta 1, rhLatent TGF-beta 1, rhTGF-beta 1.2, rhTGF-beta 2 or rhTGF-beta 3 is observed.

Blockade of Receptor-ligand Interaction

In a functional ELISA, 0.03-0.15 µg/mL of this antibody will block 50% of the binding of 50 ng/mL of Recombinant Human Neurturin (Catalog # 1297-NE) to immobilized Recombinant Human GFR alpha -2 Fc Chimera (Catalog # 613‑FR) coated at 1 µg/mL (100 µL/well). At 1 μg/mL, this antibody will block >90% of the binding.

Background: Neurturin

Neurturin is a member of the GDNF family of ligands and is distantly related to TGF-beta family ligands. It is a disulfide-linked homodimer that interacts with a receptor complex composed of a high affinity ligand binding component (GFR alpha 1 or GFR alpha 2) and a common signaling component, cRET. Neurturin promotes the survival of a variety of neurons and possibly other cell types.
