PKC theta (PRKCQ) is a calcium-independent phospholipid-dependent serine and threonine kinase that belongs to the novel PKC subfamily. PKC theta plays a critical role in several aspects of T cell biology, and knockout mice develop autoimmune disease. PKC theta functions through the activation of NF kappa B and AP1 transcription factors.

Detection of PKC theta in Jurkat Human Cell Line by Flow Cytometry.
Jurkat human acute T cell leukemia cell line was stained with Human PKC theta Monoclonal Antibody (Catalog # MAB4368, filled histogram) or isotype control antibody (Catalog # MAB003, open histogram), followed by Phycoerythrin-conjugated Anti-Mouse IgG F(ab')2Secondary Antibody (Catalog # F0102B). To facilitate intracellular staining, cells were fixed with paraformaldehyde and permeabilized with methanol.Detection of Human PKC theta by Western Blot.
Western blot shows lysates of Jurkat human acute T cell leukemia cell line. PVDF membrane was probed with 2 µg/mL of Human PKC theta Monoclonal Antibody (Catalog # MAB4368) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for PKC theta at approximately 82 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 3.Detection of Human PKC theta by Simple WesternTM.
Simple Western lane view shows lysates of Jurkat human acute T cell leukemia cell line, loaded at 0.2 mg/mL. A specific band was detected for PKC theta at approximately 82 kDa (as indicated) using 5 µg/mL of Mouse Anti-Human PKC theta Monoclonal Antibody (Catalog # MAB4368).
