Specificity

Detects human Pro Coagulation Factor II/Thrombin in direct ELISAs and Western blots. It recognizes only Pro Coagulation Factor II/Thrombin, not active Thrombin. In Western blots, less than 5% cross-reactivity with recombinant human (rh) Factor X, rhKallikrein 1, 3, 5, and 11 is observed.

Detection of Human Pro Coagulation Factor II/Thrombin by Western Blot.

Western blot shows lysates of HepG2 human hepatocellular carcinoma cell line. PVDF membrane was probed with 2 µg/mL of Mouse Anti-Human Pro Coagulation Factor II/Thrombin Monoclonal Antibody (Catalog # MAB1473) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (HAF018). A specific band was detected for Pro Coagulation Factor II/Thrombin at approximately 75 kDa (as indicated). This experiment was conducted under reducing conditions and using Western Blot Buffer Group 1.

Lyophilized from a 0.2 μm filtered solution in PBS with Trehalose. *Small pack size (SP) is supplied either lyophilized or as a 0.2 µm filtered solution in PBS.

Lyophilized product is shipped at ambient temperature. Liquid small pack size (-SP) is shipped with polar packs. Upon receipt, store immediately at the temperature recommended below.

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.

Background: Coagulation Factor II/Thrombin

Pro-Coagulation Factor II, also called Prothrombin, is a plasma serine protease central to the coagulation cascade in which it converts fibrinogen to fibrin, activates factors V, VII, VIII, XIII and forms complexes with protein C and thrombomodulin. Pro-Coagulation Factor II is an inactive precursor of thrombin that can be activated by proteolytic removal of the pro region.
