Human peripheral blood mononuclear cells (PBMCs) (A) treated with PMA and Ca2+ ionomycin overnight, or (B) resting, were stained with Human Anti-Human TNF- alpha (Research Grade Adalimumab Biosimilar) Monoclonal Antibody (Catalog # MAB9677) followed by Goat anti-Human APC-conjugated Secondary Antibody (Catalog # F0135) and Mouse anti-Human CD3 PE-conjugated Monoclonal Antibody (Catalog # FAB100P). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005). Cytotoxicity Induced by TNF-alpha and Neutralization by Human TNF-alpha Antibody.

Recombinant Human TNF-a (Catalog # 210-TA) induces cytotoxicity in the the L-929 mouse fibroblast cell line in a dose-dependent manner (orange line), as measured by Resazurin (Catalog # AR002). Cytotoxicity elicited by Recombinant Human TNF-a (0.25 ng/mL) is neutralized (green line) by increasing concentrations of Human Anti-Human TNF-a (Research Grade Adalimumab Biosimilar) Monoclonal Antibody (Catalog # MAB9677). The ND50 is typically 2-12 ng/mL in the presence of the metabolic inhibitor actinomycin D.

Measured by its ability to neutralize TNF‑ alpha -induced cytotoxicity in the L‑929 mouse fibroblast cell line. Matthews, N. and M.L. Background: TNF-alpha

Adalimumab is a biosimilar directed against the extracellular domain of TNF- alpha. This constitutes the immunogen used for the anti-Idiotype catalog numbers MAB9616 and MAB9546. The biotinylated antibody (catalog number IC9677B) can be used in flow cytometry or various assay format to measure EGFR expression and can be detected by conjugated streptavidin.
