Detection of NanoLuc® Luciferase and Large BiT by Western Blot. Western blot shows Recombinant Large BiT and Recombinant NanoLuc®. PVDF membrane was probed with 2 µg/mL of Mouse Anti-Human NanoLuc® (Nluc) Luciferase Monoclonal Antibody (Catalog # MAB10026) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for NanoLuc® and Large BiT at approximately 16 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: Luciferase
NanoLuc® (Nluc) luciferase is a small enzyme (19.1kDa) engineered for optimal performance as a luminescent reporter. NanoLuc® Binary Technology (NanoBiT™) is a two subunit system based on NanoLuc® luciferase. Large BiT (LgBiT; 17.6kDa) is the large subunit of the NanoBiT® system.
