SARS-CoV-2, which causes the global pandemic coronavirus disease 2019 (Covid-19), belongs to a family of viruses known as coronaviruses that are commonly comprised of four structural proteins: Spike protein(S), Envelope protein (E), Membrane protein (M), and Nucleocapsid protein (N) (1). SARS-CoV-2 Spike Protein (S Protein) is a glycoprotein that mediates membrane fusion and viral entry. The S protein is homotrimeric, with each ~180-kDa monomer consisting of two subunits, S1 and S2 (2). In SARS-CoV-2, as with most coronaviruses, proteolytic cleavage of the S protein into two distinct peptides, S1 and S2 subunits, is required for activation. The S1 subunit is focused on attachment of the protein to the host receptor while the S2 subunit is involved with cell fusion (3-5).

Detection of SARS-CoV-2 Spike S2 Subunit and SARS-CoV-2 Spike (Active Trimer) by Western Blot.
Western blot shows Recombinant SARS-CoV-2 Spike S2 Subunit Protein (10584-CV) and Recombinant SARS-CoV-2 Spike (Active Trimer) Protein (10549-CV). PVDF membrane was probed with 2 µg/mL of Mouse Anti-SARS-CoV-2 Spike S2 Subunit Monoclonal Antibody (Catalog # MAB10557) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (HAF018). A specific band was detected for SARS-CoV-2 Spike S2 Subunit at approximately 70 kDa (as indicated). This experiment was conducted under reducing conditions and using Western Blot Buffer Group 1.Detection of SARS-CoV-2 Spike S2 Subunit by Simple WesternTM.
Simple Western lane view shows recombinant SARS-CoV-2 Spike S1 RBD (receptor binding domain) protein (negative sample), recombinant SARS-CoV-2 Spike S1 subunit protein (negative sample), recombinant SARS-CoV-2 Spike S2 subunit protein, recombinant SARS-CoV-2 Spike S1 /S2 Subunit protein, and recombinant SARS-CoV-2 Nucleocapsid protein (negative sample), loaded at 0.2 mg/mL. Specific bands were detected for SARS-CoV-2 Spike S2 Subunit at approximately 91kDa and SARS-CoV-2 Spike S1 /S2 Subunit at approximately 230 kDa (as indicated) using 20 µg/mL of Mouse Anti-SARS-CoV-2 Spike S2 Subunit Monoclonal Antibody (Catalog # MAB10557). Q: Does SARS-CoV-2 Spike S2 Subunit Antibody, Catalog # MAB10557 cross-react with Omicron BA.1 variant? Does it cross-reaact with Omicron BA.2 variant?
A: Our testing shows that Catalog # MAB10557 cross-reacts with the Omicron BA.1 variant. We have not tested this antibody against the Omicron BA.2 variant, but given the very small difference (only 2 amino acids) between BA.1 and BA.2 in the spike S2 subunit, it is likely to cross-react with BA.2 variant as well.
