Western blot shows recombinant human Aggregan N-terminal and C-terminal fragments. PVDF membrane was probed with 2 µg/mL of Mouse Anti-Human Aggrecan Monoclonal Antibody (Catalog # MAB64892) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for the Aggrecan N-terminal fragment at approximately 75 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: Aggrecan

Aggrecan, together with type II collagen, makes up to 90% of the dry weight of healthy cartilage. It hydrates the collagen network and thus provides cartilage with its properties of compressibility and elasticity. The N-terminal region of human Aggrecan (residues 20‑675) consists of two globular domains (G1 and G2), flanking an interglobular domain (IGD). The IGD contains the cleavage sites for matrix metalloproteinases and aggrecanases.
