Detection of Human Sin1/MAPKAP1 by Western Blot.

Western blot shows lysates of human heart tissue and human kidney tissue. PVDF membrane was probed with 0.2 µg/mL of Mouse Anti-Human Sin1/MAPKAP1 Monoclonal Antibody (Catalog # MAB8168) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). Specific bands were detected for Sin1/MAPKAP1 at approximately 59-63 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: Sin1/MAPKAP1

MAPKAP1 (mitogen-activated protein kinase (MAPK) associated protein-1), also called Mip1 (MAPK interacting protein 1) or Sin1 (stress-activated protein kinase (SAPK) interacting 1), is a widely expressed 59-65 kDa cytoplasmic phosphoprotein. MAPKAP1 binds to inactive forms of signaling molecules such as MEKK2 and Ras. Alternate mRNA splicing potentially creates isoforms of 323, 330, 372, 475, 486 and 522 amino acids (aa) in humans. The human MAPKAP1 region used as an immunogen is included in all except the smallest isoform, and shares 96% aa sequence identity with mouse and rat MAPKAP1.

    Mouse white adipose tissue was homogenised and protein content was quantified by a BCA assay. Twenty micrograms of protein were resolved on a 4-12% Bis-Tris gel and transferred to nitrocellulose membranes. Membranes were probed with primary antibody MAPKAP1 diluted 1:1000 in 5% BSA 2, before incubation with anti mouse secondary horseradish peroxidase-conjugated antibody 1:5000. Blots were visualised with Immobilon Western Chemiluminescence HRP Substrate and imaged with Syngene chemiluminescence imaging system.
