TBX6 (T-box protein 6) is a 47 kDa member of the T-box family of transcriptional regulators. It is expressed during embryogenesis, plays a role in presomitic mesoderm development, and promotes delta-like 1 transcription. Human TBX6 is 436 amino acids (aa) in length. It contains one T‑box DNA-binding domain (aa 90‑277). There is one alternate splice form that shows a 15 aa substitution for aa 281‑436. Over aa 1‑280, human TBX6 shows 94% and 90% aa identity with canine and mouse TBX6, respectively.

Detection of Human TBX6 by Western Blot.
Western blot shows lysates of HT1080 human fibrosarcoma cell line and HL-60 human acute promyelocytic leukemia cell line. PVDF membrane was probed with 2 µg/mL of Goat Anti-Human TBX6 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4744) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). A specific band was detected for TBX6 at approximately 40 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 3.
TBX6 in Embryonic Mouse Mesoderm.
TBX6 was detected in immersion fixed frozen sections of embryonic mouse mesoderm (E9.5) using 10 µg/mL Human TBX6 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4744) overnight at 4 °C. Tissue was stained with the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red; Catalog # NL001) and counterstained with DAPI (blue). TBX6 in JOY6 Human iPS Cells.
TBX6 was detected in immersion fixed JOY6 human induced pluripotent stem cells undifferentiated (right panel) and differentiated into mesoderm (left panel) using Goat Anti-Human TBX6 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4744) at 0.2 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red; Catalog # NL001) and counterstained with DAPI (blue).
