Detection of CD74 in Human PBMC by Flow Cytometry. Human PBMC lymphocytes were stained with (A) Mouse Anti-Human CD74 Monoclonal Antibody (Catalog # MAB35901) or (B) Mouse IgG1 isotype control (MAB002) followed by anti-Mouse IgG APC-conjugated Secondary Antibody (F0101B) and Human CD19 PE-conjugated Monoclonal Antibody (FAB4867P). Staining was performed using our Staining Membrane-Associated Proteins protocol.

Detection of CD74 in K562 Human Cell Line by Flow Cytometry. K562 human myelogenous leukemia cell line was stained with Mouse Anti-Human CD74 Monoclonal Antibody (Catalog # MAB35901, filled histogram) or Mouse IgG1 isotype control (MAB002,open histogram) followed by anti-Mouse IgG APC-conjugated Secondary Antibody (F0101B). Staining was performed using our Staining Membrane-Associated Proteins protocol.

Detection of CD74 in Human Blood Cells by Flow Cytometry. Human peripheral blood lymphocytes were labeled with Mouse Anti-Human CD74 Monoclonal Antibody (Catalog # MAB35901) followed by Phycoerythrin-conjugated Anti-Mouse IgG F(ab')2Secondary Antibody (Catalog # F0102B) and Human CD19 APC-conjugated Monoclonal Antibody (Catalog # FAB4867A). Quadrant markers were set based on control antibody staining (Catalog # MAB002).

Background: CD74
CD74 is a transmembrane protein which, on antigen presenting cells, is the invariant chain that chaperones MHC class II dimers from the endoplasmic reticulum to the cell surface. The cytoplasmic portion of CD74 can be released by intramembrane cleavage and activate NF kappa B. CD74 is up-regulated in several cancers and is also expressed by non-immune cells during inflammation. This antibody was raised to a splice variant of human CD74 which lacks the Thyroglobulin type 1 repeat in the extracellular domain. Within comparable regions, human CD74 shares 70% and 73% amino acid sequence identity with mouse and rat CD74, respectively.
