Detection of CD74 in Human Blood Cells by Flow Cytometry. Human peripheral blood lymphocytes were labeled with Mouse Anti-Human CD74 Monoclonal Antibody (Catalog # MAB35901) followed by Phycoerythrin-conjugated Anti-Mouse IgG F(ab')2 Secondary Antibody (Catalog # F0102B) and Human CD19 APC-conjugated Monoclonal Antibody (Catalog # FAB4867A). Quadrant markers were set based on control antibody staining (Catalog # MAB002).

CD74 is a transmembrane protein which, on antigen presenting cells, is the invariant chain that chaperones MHC class II dimers from the endoplasmic reticulum to the cell surface. The cytoplasmic portion of CD74 can be released by intramembrane cleavage and activate NF kappa B. CD74 is up-regulated in several cancers and is also expressed by non-immune cells during inflammation. This antibody was raised to a splice variant of human CD74 which lacks the Thyroglobulin type 1 repeat in the extracellular domain. Within comparable regions, human CD74 shares 70% and 73% amino acid sequence identity with mouse and rat CD74, respectively.
