IFN‑ beta Inhibition of EMCV-induced Cytopathy and Neutralization by Human IFN‑ beta Antibody.
Recombinant Human IFN-beta (8499-IF) reduces the Encephalomyocarditis Virus (EMCV)-induced cytopathy in the HeLa human cervical epithelial carcinoma cell line in a dose-dependent manner (orange line), as measured by crystal violet staining. Inhibition of EMCV activity elicited by Recombinant Human IFN-beta (50 pg/mL) is neutralized (green line) by increasing concentrations of Human IFN-beta Monoclonal Antibody (Catalog # MAB814). The ND50 is typically 1.50-18.0 µg/mL.Detection of IFN‑ beta in M1 macrophages by Flow Cytometry.
M1 macrophages treated with 50 ng/mL each of rhGM-CSF (215-GM) for 6 days followed by an overnight polarization with IFNg and LPS (285-IF) and restimulated with monensin for 3 hours (3uM) were stained with Mouse Anti-Human CD14 PE‑conjugated Monoclonal Antibody (Catalog # FAB3832P) and either (A) Mouse Anti-Human IFN‑ beta Monoclonal Antibody (Catalog # MAB814) or (B) Mouse IgG1 Isotype Control (Catalog # MAB002) followed by Allophycocyanin-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # F0101B).
