Detection of Human and Mouse SOCS‑3 by Western Blot.
Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line, Jurkat human acute T cell leukemia cell line, U937 human histiocytic lymphoma cell line, and C2C12 mouse myoblast cell line. PVDF membrane was probed with 0.1 µg/mL of Mouse Anti-Human/Mouse SOCS‑3 Monoclonal Antibody (Catalog # MAB5696) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for SOCS‑3 at approximately 30 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
SOCS‑3 in U2OS Human Cell Line.
SOCS‑3 was detected in immersion fixed U2OS human osteosarcoma cell line using Mouse Anti-Human/Mouse SOCS‑3 Monoclonal Antibody (Catalog # MAB5696) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 493-conjugated Anti-Mouse IgG Secondary Antibody (green; Catalog # NL009) and counterstained with DAPI (blue). Suppressor of cytokine signaling 3 (SOCS-3), also known as CIS3, is a widely expressed 30 kDa protein that inhibits signal transduction induced by multiple cytokines including Erythropoietin, IL-6, LIF, Oncostatin M, and Leptin. It binds downstream signaling molecules such as JAK kinases and targets them for ubiquitin-mediated degradation, essential in a negative feedback control loop. Human SOCS-3 shares 96% aa sequence identity with mouse and rat SOCS-3.
