Rat CXCL3/CINC-2 alpha / beta Antibody Summary

Species Reactivity
Detects rat CXCL3/CINC-2 alpha / beta in ELISAs and Western blots. In sandwich immunoassays, 27.9% cross‑reactivity with recombinant rat (rr) CINC-3 is observed and no cross-reactivity or interference with rrCINC‑1, recombinant mouse (rm) KC, rmMIP-2,  or recombinant human GRO alpha is observed.
Monoclonal Mouse IgG2B Clone # 83504
Protein A or G purified from ascites
E. coli-derived recombinant rat CXCL3/CINC-2 alpha / beta
Accession # Q10746
Lyophilized from a 0.2 μm filtered solution in PBS with Trehalose. *Small pack size (SP) is supplied either lyophilized or as a 0.2 µm filtered solution in PBS.
Endotoxin Level
<0.10 EU per 1 μg of the antibody by the LAL method.

Recombinant Rat CXCL3/CINC-2 alpha (Catalog # 516-CA)
Recombinant Rat CXCL3/CINC-2 beta (Catalog # 517-CB)

Rat CXCL3/CINC-2 alpha / beta Sandwich Immunoassay

Recommended Concentration
ELISA Capture (Matched Antibody Pair)
2-8 µg/mL

Use in combination with:

Detection Reagent: Rat CXCL3/GRO gamma/CINC-2/DCIP-1 Biotinylated Antibody (Catalog # BAF516)

Standard: Recombinant Rat CXCL3/CINC-2 beta Protein (Catalog # 517-CB)

Standard: Recombinant Rat CXCL3/CINC-2 alpha Protein (Catalog # 516-CA)

Measured by its ability to neutralize CXCL3/CINC‑2 alpha -induced chemotaxis in the BaF3 mouse pro‑B cell line transfected with human CXCR2. The Neutralization Dose (ND50) is typically 0.5-2.5 µg/mL in the presence of 0.05 µg/mL Recombinant Rat CXCL3/CINC‑2 alpha.

Please Note: Optimal dilutions should be determined by each laboratory for each application. General Protocols are available in the Technical Information section on our website.

Chemotaxis Induced by CXCL3/CINC‑2 alpha and Neutralization by Rat CXCL3/CINC‑2 alpha / beta Antibody. Recombinant Rat CXCL3/ CINC-2a (Catalog # 516-CA) chemoattracts the BaF3 mouse pro-B cell line transfected with human CXCR2 in a dose-dependent manner (orange line). The amount of cells that migrated through to the lower chemotaxis chamber was measured by Resazurin (Catalog # AR002). Chemotaxis elicited by Recombinant Rat CXCL3/ CINC-2a (0.05 µg/mL) is neutralized (green line) by increasing concentrations of Rat CXCL3/CINC-2a/ beta Monoclonal Antibody (Catalog # MAB516). The ND50 is typically 0.5-2.5 µg/mL.

Background: CXCL3/GRO gamma/CINC-2/DCIP-1

CINC-2 alpha and -2 beta are mRNA splice variant isoforms that differ by only 3 amino acids at their amino-termini. These proteins are CXC chemokines most closely related to human GROs and mouse MIP-2. They are produced by granulation tissue, activated macrophages and fibroblasts. CINC-2 biological activities are mediated by CXCR1 or CXCR2.
