Detection of Human AG‑3 by Western Blot.

Western blot shows lysates of T47D human breast cancer cell line. PVDF membrane was probed with 1 µg/mL of Mouse Anti-Human AG-3 Monoclonal Antibody (Catalog # MAB6307) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for AG-3 at approximately 16 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

AG‑3 in Human Breast.

AG-3 was detected in immersion fixed paraffin-embedded sections of human breast using Mouse Anti-Human AG-3 Monoclonal Antibody (Catalog # MAB6307) at 15 µg/mL overnight at 4 °C. Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using the Anti-Mouse HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS002) and counter-stained with hematoxylin (blue). Background: AG-3

AG-3 (Anterior gradient protein 3; also known as AGR3 and BCMP11) is a 17 kDa (predicted) member of the AGR family of proteins. It is expressed by intestinal epithelial and breast cancer cells. AG-3 is presumably secreted and binds to both alpha -dystroglycan and C4.4a/LYPD3. Mature human AG-3 is 145 amino acids (aa) in length (aa 22‑166). Over aa 22-166, human AG-3 shares 92% amino acid sequence identity with mouse AG-3.
