Detection of Human AG‑3 by Western Blot.
Western blot shows lysates of T47D human breast cancer cell line. PVDF membrane was probed with 1 µg/mL of Mouse Anti-Human AG‑3 Monoclonal Antibody (Catalog # MAB6307) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for AG‑3 at approximately 16 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
AG‑3 in Human Breast.
AG‑3 was detected in immersion fixed paraffin-embedded sections of human breast using Mouse Anti‑Human AG‑3 Monoclonal Antibody (Catalog # MAB6307) at 15 µg/mL overnight at 4 °C. Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using the Anti-Mouse HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS002) and counterstained with hematoxylin (blue). AG-3 (Anterior gradient protein 3; also known as AGR3 and BCMP11) is a 17 kDa (predicted) member of the AGR family of proteins. It is expressed by intestinal epithelial and breast cancer cells. AG-3 is presumably secreted and binds to both alpha -dystroglycan and C4.4a/LYPD3. Mature human AG-3 is 145 amino acids (aa) in length (aa 22‑166). Over aa 22-166, human AG-3 shares 92% amino acid sequence identity with mouse AG-3.
