Human gp130 ELISA Standard Curve.
Recombinant Human gp130 protein was serially diluted 2-fold and captured by Mouse Anti-Human gp130 Monoclonal Antibody (Catalog # MAB228) coated on a Clear Polystyrene Microplate (Catalog # DY990). Mouse Anti-Human gp130 Monoclonal Antibody (Catalog # MAB2281) was biotinylated and incubated with the protein captured on the plate. Detection of the standard curve was achieved by incubating Streptavidin-HRP (Catalog # DY998) followed by Substrate Solution (Catalog # DY999) and stopping the enzymatic reaction with Stop Solution (Catalog # DY994).Detection of gp130 by Western Blot
gp130 and IL-6Ra expression in SCC-4 cells and DOK cells. DOK and SCC-4 cells were untreated or treated with rhIL-6 (30 ng/ml) for 24 h. Lysates were collected and samples were run on 8% SDS-PAGE gel. Western Blot were probed with anti-gp130 and anti-IL-6Ra antibody with anti-beta -actin used as a loading control. Experiments was done in triplicate. Densitometric analysis was performed using image lab software on gp130, IL-6Ra and beta -actin blots. Sample blots for (A) DOK and (B) SCC-4 and collated densitometry data for (Fig. S5A) gp130 and (Fig. S5B) IL-6Ra are presented. Results were plotted using Graphpad prism 8. Values represent the mean ± S.E.M of three independent experiments. Statistical analysis was performed using one-way ANOVA with a post hoc Sidak’s test to compare mean values among the groups, *p < 0.05; **p < 0.01; ***p < 0.001.
