Detection of Human HSP47 by Western Blot.
Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line, HepG2 human hepatocellular carcinoma cell line, A549 human lung carcinoma cell line, and OVCAR-3 human ovarian carcinoma cell line. PVDF membrane was probed with 0.1 µg/mL of Mouse Anti-Human HSP47 Monoclonal Antibody (Catalog # MAB91661) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for HSP47 at approximately 47 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.HSP47 in BG01V Human Embryonic Stem Cells.
HSP47 was detected in immersion fixed BG01V human embryonic stem cells differentiated to hepatocytes using Mouse Anti-Human HSP47 Monoclonal Antibody (Catalog # MAB91661) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm. HSP47 was detected in immersion fixed paraffin-embedded sections of human breast cancer tissue using Mouse Anti-Human HSP47 Monoclonal Antibody (Catalog # MAB91661) at 25 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Mouse HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS002) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm.
