Please Note: Optimal dilutions should be determined by each laboratory for each application. General Protocols are available in the Technical Information section on our website.
Detection of Human IKK epsilon by Western Blot. Western blot shows lysates of MDA-MB-468 human breast cancer cell line, Raji human Burkitt's lymphoma cell line, and Ramos human Burkitt's lymphoma cell line. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human IKKe Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3199) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). A specific band was detected for IKKe at approximately 80 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
IKK epsilon in Ramos Human Cell Line. IKKe was detected in immersion fixed Ramos human Burkitt's lymphoma cell line using Goat Anti-Human IKKe Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3199) at 15 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red; Catalog # NL001) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm. View our protocol for Fluorescent ICC Staining of Non-adherent Cells.

Background: IKK epsilon
I kappa B kinase epsilon (IKK epsilon ) is also known as IKBKE and IKKi. The classical active IKK complex, composed of IKK alpha, IKK beta, and two forms of processed IKK gamma, phosphorylates and inactivates I kappa B, resulting in the release and nuclear translocation of active NF-kappa B. IKK epsilon is an IKK-related kinase with 30% structural homology to IKK alpha and beta, and is present in a subset of IKK complexes. IKK epsilon activation has been implicated in the initiation of anti-viral gene expression and in the pathogenesis of breast cancer.
