Western blot shows lysates of human, mouse, and rat heart tissue. PVDF membrane was probed with 0.1 µg/mL of Mouse Anti-Human/Mouse/ Rat a-Cardiac Actin Monoclonal Antibody (Catalog # MAB9308) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for a-Cardiac Actin at approximately 42 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Specific staining was localized to cytoplasm.

Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm of cardiomyocytes. A specific band was detected for alpha -Cardiac Actin at approximately 50 kDa (as indicated) using 2 µg/mL of Mouse Anti-Human/Mouse/Rat alpha -Cardiac Actin Monoclonal Antibody (Catalog # MAB9308). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

Background: alpha-Cardiac Actin

Actin, alpha cardiac muscle 1 (ACTC1) is a 377 aa cytoskeletal filament in cardiac muscle which interacts with the 50-KDa domain of the myosin motor domain in cardiac muscle contraction. Actin is highly conserved across species, and differs from skeletal muscle actin (ACTA1) by only four amino acids. Mutations in the ACTC1 gene are linked to familial hypertrophy cardiomyopathy, atrial septal defects, arrhythmia, and chronic inflammatory cardiomyopathy.
