MMP‑8 in Human Breast Cancer Tissue.

MMP-8 was detected in immersion fixed paraffin-embedded sections of human breast cancer tissue using 8 µg/mL Mouse Anti-Human MMP-8 Monoclonal Antibody (Catalog # MAB9081) overnight at 4 °C. Tissue was stained with the Anti-Mouse HRP-DAB Cell & Tissue Staining Kit (brown; CTS002) and counterstained with hematoxylin (blue). Specific labeling was localized to epithelial cells in terminal ductules (round) and intralobular duct (elongated) composing glandular lobules. Human MMP-8 ELISA Standard Curve.

Recombinant Human MMP‑8 protein was serially diluted 2-fold and captured by Mouse Anti-Human MMP‑8 Monoclonal Antibody (MAB908) coated on a Clear Polystyrene Microplate (DY990). Mouse Anti-Human MMP‑8 Monoclonal Antibody (Catalog # MAB9081) was biotinylated and incubated with the protein captured on the plate. Detection of the standard curve was achieved by incubating Streptavidin-HRP (DY998) followed by Substrate Solution (DY999) and stopping the enzymatic reaction with Stop Solution (DY994).

Detection of MMP‑8 in Jurkat cells by Flow Cytometry

Jurkat cells were stained with Mouse Anti-Human MMP‑8 Monoclonal Antibody (Catalog # MAB9081, filled histogram) or isotype control antibody (Catalog # MAB003, open histogram) followed by Phycoerythrin-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # F0102B). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005). Background: MMP-8

Matrix metalloproteinases (MMPs) are a family of zinc and calcium dependent endopeptidases with the combined ability to degrade all the components of the extracellular matrix. MMP-8 (neutrophil collagenase) is expressed in neutrophils, where it is stored in specific granules. MMP-8 release from the neutrophils is stimulated by various factors such as interleukins 1 and 8, TNF-alpha and GM-CSF. MMP-8 is capable of cleaving types I, II and III triple-helical collagen, gelatin peptides, fibronectin, proteoglycans, aggrecan, serpins, beta -casein and peptides such as angiotensin and substance P. In addition to its function in phagocytosis,
MMP‑8 has a high capacity for infiltrating connective tissue, and is implicated in the breakdown of the extracellular matrix in diseases such as rheumatoid arthritis. Structurally, MMP-8 consists of several domains: a pro-domain that is cleaved upon activation, a catalytic domain containing the zinc-binding site, a short hinge region and a hemopexin-like domain. MMP-8 is heavily glycosylated.
