    Measured by its ability to neutralize CCL4/MIP‑1 beta -induced chemotaxis in the BaF3 mouse pro‑B cell line transfected with human CCR5. The Neutralization Dose (ND50) is typically 0.3-1.2 µg/mL in the presence of 0.04 µg/mL Recombinant Human CCL4/MIP‑1 beta.

Chemotaxis Induced by CCL4/MIP‑1 beta and Neutralization by Human CCL4/MIP‑1 beta Antibody. Recombinant Human
CCL4/MIP‑1 beta (Catalog #
271-BME) chemoattracts the BaF3 mouse pro‑B cell line transfected with human CCR5 in a dose-dependent manner (orange line). The amount of cells that migrated through to the lower chemotaxis chamber was measured by Resazurin (Catalog # AR002). Chemotaxis elicited by Recombinant Human
CCL4/MIP‑1 beta (0.04 µg/mL) is neutralized (green line) by increasing concentrations of Mouse Anti-Human
CCL4/MIP‑1 beta Monoclonal Antibody (Catalog # MAB271). The ND50 is typically
0.3-1.2 µg/mL.
Intracellular Staining by Flow Cytometry
Detection of CCL4/MIP‑1 beta in Human Blood Monocytes by Flow Cytometry. Human peripheral blood monoctyes treated with LPS and Monensin for 5 hours were stained with Mouse Anti-Human CD14 APC‑conjugated Monoclonal Antibody (Catalog # FAB3832A) and either (A) Mouse Anti-Human CCL4/MIP‑1 beta Monoclonal Antibody (Catalog # MAB271) or (B) Mouse IgG2B Flow Cytometry Isotype Control (Catalog # MAB0041) followed by Allophycocyanin-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # F0101B).To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005). Background: CCL4/MIP-1 beta
CCL4, also known as Macrophage Inflammatory Protein 1 beta (MIP-1 beta ) is a member of the CC or beta chemokine subfamily. CCL4 is expressed primarily by T cells, B cells, and monocytes after antigen or mitogen stimulation. The functional receptor for CCL4 has been identified as CCR5. Mature human CCL4 shares 77% and 80% aa sequence identity with mouse and rat CCL4, respectively.
