Oct-4A (red) & E-Cadherin (green) in iPS2 human induced pluripotent stem cells. Oct-4A and E-Cadherin were detected in iPS2 human induced pluripotent stem cells using a Mouse Anti-Human Oct-4A Monoclonal Antibody (Catalog # MAB17591) and a Goat Anti-Human E-Cadherin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF648). The cells were stained for Oct-4A using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007), and stained for E-Cadherin using the NorthernLights™ 493-conjugated Anti-Goat IgG Secondary Antibody (green; Catalog # NL003). Specific staining of Oct-4A in the nucleus and E-Cadherin at the plasma membrane suggests this population of cells is pluripotent. CD90/Thy1 & CD45 in human mesenchymal stem cells. Human mesenchymal stem cells (MSC) were stained with a PE-conjugated Mouse Anti-Human CD90/Thy1 Monoclonal Antibody (Catalog # FAB1430P) and an APC-conjugated Mouse Anti-Human CD45 Monoclonal Antibody (Catalog # FAB2067A). Gate based on control antibodies. The data show high levels of CD90/Thy1 and low expression of CD45, a phenotype that is consistent with a multipotent MSC starting population.
SSEA-1 (red) & SSEA-4 (green) in live iPS2 human induced pluripotent stem cells. iPS2 human induced pluripotent stem cells were grown on irradiated mouse embryonic fibroblasts (Catalog # PSC001) and stained using the NL493-conjugated SSEA-4 (green) and NL557-conjugated SSEA-1 (red) antibodies included in the Human Pluripotent Stem Cell Live Cell Imaging Kit (Catalog # SC023). The cells were counterstained with Hoechst 33342 (blue). The colonies are positive for the stem cell marker SSEA-4 and are negative for SSEA-1, suggesting that these colonies primarily contain undifferentiated human stem cells. This kit also contains a NL557-conjugated Mouse Anti-Human TRA-1-60(R) Monoclonal Antibody.
