Cytokine ELISA Kits and Multiplex Immunoassays: Frequently Asked Questions

A cytokine ELISA (enzyme-linked immunosorbent assay) is a highly sensitive and specific immunoassay for quantitatively measuring individual cytokine proteins in biological samples such as serum, plasma, cell culture supernatants, or tissue homogenates. This assay uses antibodies that specifically bind the target cytokine to accurately determine its concentration. The most common format, the sandwich ELISA, involves coating a microplate with capture antibody, adding the sample to immobilize the cytokine, then applying a conjugated detection antibody. Following the addition of a substrate, a signal is generated that is proportional to the amount of cytokine present in the sample. Cytokine ELISAs are essential for studying cytokines expression profiles, understanding immune responses, and evaluating the effects of experimental treatments or disease states. 

Both ELISA and Luminex Assays are antibody-based immunoassays used to quantify proteins such as cytokines, chemokines, growth factors, and biomarkers. The primary difference is that an ELISA measures one analyte per well, whereas a Luminex Assay enables the simultaneous measurement of multiple analytes in a single sample. 

  • An ELISA uses antibodies immobilized on a microplate to capture a target analyte. Detection is typically achieved with a conjugated detection antibody and a substrate that produces a colorimetric signal proportional to the analyte concentration. 
  • Luminex Assays use color-coded superparamagnetic beads, with each bead set coated with an analyte-specific capture antibody. Multiple bead populations are combined in the same reaction and incubated with the sample, allowing simultaneous detection of up to 50 analytes in a single sample. Captured analytes are subsequently detected using a cocktail of biotinylated detection antibodies and a streptavidin-phycoerythrin (PE) conjugated. Using a Luminex instrument, beads are excited by one laser to determine the bead region and corresponding assigned analyte. Another laser determines the magnitude of the PE-derived signal, which is proportional to the amount of analyte bound. Multiple readings are taken at each bead region, ensuring robust detection.

ELISA Kits and multiplex immunoassays are available for hundreds of cytokines and other immune-related analytes, including interleukins, interferons, tumor necrosis factors, chemokines, colony-stimulating factors, and growth factors. Common targets include:

  • Pro-inflammatory markers such as IL-1α, IL-1β, IL-6, IL-17A, IL-18, GM-CSF, TNF-α, and IFN-γ
  • Anti-inflammatory cytokines such as IL-10, IL-1ra, and TGF-β
  • Cytokines important for adaptive immunity and chemokines including IL-2, IL-4, IL-5, IL-7, IL-15, IL-22, IL-23, CCL2, CCL3, CCL4, CCL5, and CXCL10.

The number of analytes that can be measured in a multiplex immunoassay depends on the platform, assay design, and biological matrix. Luminex assays, by comparison, offer significantly greater multiplexing capacity, enabling the simultaneous measurement of up to 50 analytes in a single sample. This makes Luminex technology particularly well-suited for cytokine profiling, immune monitoring, and biomarker discovery studies where a broader view of biological pathways is needed. By generating data on multiple targets in a single run, multiplex immunoassays help conserve valuable sample volume, reduce time and costs, and provide a more comprehensive understanding of complex biological processes.
