Detection of SARS-CoV-2 Spike RBD by ELISA. Recombinant SARS-CoV-2 Spike RBD protein was serially diluted 2-fold and captured by a Mouse Anti-SARS-CoV-2 Spike RBD Monoclonal Antibody (Catalog # MAB10580) coated on a Clear Polystyrene Microplate (Catalog # DY990). A different Mouse Anti-SARS-CoV-2 Spike RBD Monoclonal Antibody (Catalog # MAB105406) was biotinylated and used to detect the captured SARS-CoV-2 Spike RBD protein. Detection of the standard curve was achieved by adding Streptavidin-HRP (Catalog # DY998) to the plate, followed by a Substrate Solution (Catalog # DY999) and then a Stop Solution (Catalog # DY994) to stop the enzymatic reaction.

Detection of SARS-CoV-2 Spike Protein by Flow Cytometry. HEK293 human embryonic kidney cell line, transfected with human ACE-2 and eGFP, was incubated with Recombinant SARS-CoV-2 Spike S1 Subunit His-Tag Protein (Catalog # 10522-CV), then stained with a Mouse Anti-SARS-CoV-2 Spike RBD Monoclonal Antibody (A; Catalog # MAB105403) or a Mouse IgG1 Isotype Control Antibody (B; Catalog # MAB002), followed by an APC-Conjugated Goat Anti-Mouse IgG Secondary Antibody (Catalog # F0101B). SARS-CoV-2 Spike protein, bound to ACE-2 expressed on the surface of the transfected HEK293 cells, was detected.

Spike RBD was detected in immersion-fixed, paraffin-embedded sections of SARS-CoV-2 infected human lung tissue using a Mouse Anti-SARS-CoV-2 Spike RBD Monoclonal Antibody (Catalog # MAB105802), followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC001). Prior to incubation with the primary antibody, the tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). The tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to SARS-CoV-2 infected cells.

The PVDF membrane was probed with a Mouse Anti-SARS-CoV-2 Spike S2 Subunit Monoclonal Antibody (Catalog # MAB10557) followed by an HRP-Conjugated Donkey Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for SARS-CoV-2 Spike S2 Subunit at approximately 70 kDa (as indicated).
